The alleged assault occurred in the summer that Ronaldo left Manchester United for Real Madrid to become the then most expensive player in the world.
Ms Mayorga claims to have originally signed a non-disclosure agreement with Ronaldo's lawyers worth $375,000 (£289,000) but is now seeking damages against Ronaldo and his legal team.
He made the statement on an Instagram Live video. They want to be famous, to say my name. In an Instagram video, Ronaldo dismissed the claim and said: "They want to promote by my name". Yeah but it's part of the job. "I'm [a] happy man and all, all good".
The woman, Ms Kathryn Mayorga, 34, alleges that the rape took place in June 2009 after they met at the Palms hotel and casino. But an earlier version allegedly had a written admission where it said "she said no and stop several times".
But while she was changing, Ronaldo, 33, entered the room, exposed himself and asked her to perform a sex act.
She says he apologized after it was over, "stating he was sorry, he was usually a gentleman".

Lindsay Lohan Punched by Mother After Trying to Take Her Son
At least, we can all stop remembering her turns in The Parent Trap and Mean Girls fondly, because she's just a bad person. Lindsay Lohan isn't recognized for any type of humanitarian work, but the actress seems to have human rights on her mind.
According to Las Vegas Metropolitan Police Department spokesman Aden Ocampo-Gomez, Mayorga did not provide an assailant's name or location of the alleged assault in her 2009 complaint. Schertz says they will sue Der Spiegel although did not follow through with similar threats when the magazine first reported the non-disclosure agreement previous year .
Contacted late on Monday, Ronaldo's representatives did not immediately return a request for comment about the reopening of the case by Las Vegas police.
Gestifute did not answer any specific questions about the content of the report.
Der Spiegel's deputy editor-in-chief, Alfred Weinzierl, on Sunday said the magazine stood fully by its story.
Kathryn has a degree in journalism from the University of Nevada, but didn't work in the field. We say: "It is allowed under Germany's press law", Weinzierl said in a statement to Reuters.
The unidentified "fixers" were also named as co-defendants in the lawsuit, which accuses the Juventus forward of violating the nondisclosure agreement.
Cristiano Ronaldo's rape allegations were first reported in a German newspaper last April but - after a vehement denial - just kind of went away. Last year, German magazine Der Spiegel reported on the settlement documents.
